AMP production

What is AMP production?
AMP production testing measures how cosmetic ingredients affect endogenous antimicrobial peptide production in skin models. Gene expression, peptide levels, and suitable functional assays help investigate innate defense responses without assuming that increased production is always beneficial.
Why do you need this service?
AMP production studies investigate whether an ingredient changes endogenous antimicrobial peptide responses in skin models. Interpret peptide expression or abundance alongside viability and, where relevant, a separately measured functional response.

AMP production
Antimicrobial peptides are components of the skin’s innate defense system. In this testing service, AMP production refers to the response of skin cells to an ingredient or formulation, rather than the commercial synthesis of peptide raw materials.
Measuring the skin-model response
Labs use keratinocyte cultures or reconstructed skin models to examine changes after treatment and, where relevant, a defined challenge. The selected peptide family and model determine the measurements.
- Expression of genes encoding antimicrobial peptides
- Peptide concentrations in cells or culture medium
- Concentration- and time-dependent responses
- Cell viability and inflammatory markers alongside AMP measurements
Expression data and protein measurements address different stages of production and should not be treated as equivalent.
Functional testing and controls
Where the research question requires it, functional assays can investigate antimicrobial activity against selected organisms. Purified or synthesized reference peptides may serve as controls, but making them is an assay-support activity rather than the core service.
Reports should explain whether treatment changed peptide expression, measured peptide abundance, or functional activity. These findings can support research into skin defense, but do not prove infection prevention, broad pathogen protection, or improved barrier function without additional evidence.
Investigating host antimicrobial peptide responses
Choose measurements that distinguish a change in transcript expression from a change in peptide abundance or activity. A defined challenge can be included where it fits the research question and model. Synthesizing an antimicrobial peptide is a different service from measuring the model’s own peptide production.
Changes in cytokines can provide context but do not substitute for the AMP endpoint. Increased peptide levels are not automatically beneficial, and lower signals can reflect toxicity or model limitations. The report should identify which peptides and responses were assessed.
These studies can inform innate-defence research and formulation development. They do not establish broad pathogen protection, finished-product preservation or a clinical skin-barrier benefit. Any microbial inhibition or human-use outcome needs suitable supporting evidence.
