NMF production

What is NMF production?
NMF production testing examines whether ingredients support formation of natural moisturizing factor components in epidermal models. Analysis of amino acids and related water-binding compounds helps distinguish endogenous production from ingredients deposited by a formulation.
Why do you need this service?
NMF studies compare selected water-binding components and their formation in epidermal models. Distinguish endogenous production from supplied ingredients, altered breakdown and retention before relating the result to hydration.

NMF production
Natural moisturizing factor (NMF) is a mixture of water-binding substances in the stratum corneum. Many components derive from filaggrin breakdown, while others have different sources. This service focuses on their formation in skin-relevant models rather than compositional analysis of a moisturizer alone.
Component analysis
Differentiated epidermal models or suitable tissue samples can be analyzed by chromatographic methods for selected components. Sampling, extraction, and normalization should be defined consistently across treatments.
- Free amino acid profiles
- Pyrrolidone carboxylic acid and urocanic acid where relevant
- Other selected compounds such as lactate or urea
- Filaggrin expression and processing as supporting endpoints
Distinguishing production from replenishment
A formula containing amino acids or other NMF constituents can increase measured amounts without stimulating endogenous production. Appropriate controls and model design are therefore essential. NMF abundance may also reflect altered breakdown or retention. Functional hydration and barrier measurements can complement the analysis, but a higher component concentration alone does not establish improved barrier recovery or sustained moisturization on human skin.
Following NMF components
Chromatographic analysis can compare free amino acids, pyrrolidone carboxylic acid, urocanic acid and other relevant components. Filaggrin expression and processing provide supporting information about an important source of NMF. Ceramides are barrier lipids, not NMF components.
Control sampling, extraction and normalisation across treatments. Formulas containing amino acids or other measured compounds may raise recovered amounts by deposition. A study needs a design that can distinguish replenishment from endogenous production.
Use the results to refine a hydration-related hypothesis and compare candidates. A higher component level does not automatically establish improved barrier recovery, sustained moisturisation, formula stability or a safe use concentration. Human performance claims require relevant outcome evidence.
