MMP inhibition

What is MMP inhibition?
MMP inhibition testing assesses whether cosmetic ingredients reduce the activity or production of matrix metalloproteinases, enzymes involved in extracellular matrix breakdown. Biochemical and skin cell assays distinguish direct enzyme inhibition from changes in MMP expression.
Why do you need this service?
MMP studies compare enzyme activity or production associated with extracellular-matrix breakdown. Use them to distinguish direct biochemical inhibition from altered expression in a skin-cell model.

MMP inhibition testing
Matrix metalloproteinases contribute to the turnover of collagen and other extracellular matrix components. Testing investigates whether an ingredient inhibits selected MMPs directly or affects their production in skin cells, helping assess a proposed matrix-protective mechanism.
Enzyme activity assays
Fluorogenic substrate assays expose purified enzymes to a test material and measure substrate breakdown. Panels may include MMP-1, MMP-2 or MMP-9, depending on the target. A concentration-response series can yield an IC50 when the data support its calculation.
- Concentration-response comparisons
- Time-dependent inhibition measurements
- Selectivity across different MMPs
- Activity under defined pH conditions
Controls should account for fluorescence interference and other effects of the test material that could be mistaken for enzyme inhibition.
Cellular expression and secretion
Fibroblast or keratinocyte studies measure how ingredients influence MMP gene expression and protein secretion. Methods include qPCR for MMP-1, MMP-3 or MMP-13, ELISA for secreted proteins and western blotting for protein expression.
Expression and protein abundance are not equivalent to enzyme activity, so complementary activity measurements may be needed. Cytotoxicity screening helps interpret reduced signals. These results describe effects in the selected model and do not alone demonstrate prevention of collagen loss or wrinkles in human skin.
Investigating matrix metalloproteinase responses
Choose the MMP and substrate for the proposed question. Different MMPs have different substrate profiles; a gelatinase assay should not be described as a general elastase test. MMP-3 work can address selected matrix substrates or pro-MMP activation rather than assuming direct degradation of native fibrillar collagen III.
A cell-based study may examine production or release after a defined stimulus. Those results are distinct from direct enzyme inhibition. Viability, analytical interference and substrate suitability should inform interpretation.
The findings can support matrix-mechanism research and candidate comparisons. There is no universal conversion from an inhibition percentage to human firmness, wrinkle reduction or clinically improved skin structure.
