Keratin synthesis

What is Keratin synthesis?
Keratin synthesis testing measures how ingredients influence keratin production in living hair follicle cells or suitable follicle models. It investigates protein formation during hair development, not repair by new synthesis in an existing hair shaft.
Why do you need this service?
Keratin studies compare responses in living follicular cells as new hair forms. Existing hair fibres cannot synthesise protein, so strengthening or repair claims for emerged hair need direct fibre-performance testing.

Keratin synthesis
Hair keratins are produced by living follicular cells as the hair shaft forms. The emerged fiber is nonliving and cannot synthesize proteins, so this service is biologically distinct from coating damaged hair or reducing protein loss during styling.
Gene and protein analysis
Follicular keratinocyte cultures or isolated follicle models can be used to measure changes in selected hair keratins. Quantitative reverse-transcription PCR assesses gene expression, while Western blotting or immunofluorescence provides protein-related evidence.
- Hair keratin targets appropriate to the model, such as K31, K85, or K86
- Concentration-response and time-course comparisons
- Protein abundance or deposition alongside gene expression
- Viability and differentiation markers
Relating production to hair properties
Increased keratin mRNA alone does not demonstrate greater protein synthesis, stronger fibers, or faster growth. Protein measurements and follicle observations can strengthen interpretation, while mechanical testing on hair fibers addresses strength directly. The model should also distinguish changes in cell number from changes in keratin production per cell. Findings support mechanism research and ingredient comparison rather than proof of repaired existing hair.
Investigating keratin formation
Follicular keratinocyte or isolated-follicle models can compare selected hair-keratin transcripts, protein abundance and deposition. RT-PCR, Western blotting and immunofluorescence provide different levels of evidence. Viability, differentiation and cell-number measurements help interpret the response.
Increased mRNA alone does not establish greater protein synthesis or stronger hair. Follicle observations and protein measurements can support mechanism research, while mechanical tests on fibres address strength directly.
Keep existing-fibre effects separate
Conditioners, hydrolysed proteins and coatings may change the properties of damaged fibres without inducing new keratin synthesis. Compare those effects using suitable fibre methods. A follicular assay cannot demonstrate rebuilding of nonliving hair or establish a universal strengthening percentage or treatment period.
