Proteasome activity

What is Proteasome activity?
Proteasome activity testing measures protein-degradation activity in skin-cell or biochemical models after ingredient exposure. Fluorogenic assays and complementary protein-damage measurements help investigate cellular protein maintenance under defined stress conditions.
Why do you need this service?
Proteasome studies help investigate protein-degradation activity after ingredient exposure in skin-cell or biochemical models. Select the assay for the proteasome activity of interest and interpret it alongside cell condition.

Proteasome activity
The proteasome participates in the degradation of intracellular proteins, including some damaged proteins. This service investigates how a cosmetic ingredient affects selected proteasome activities, often in research involving oxidative or pollutant stress.
Activity and protein-damage measurements
Labs may use fluorogenic substrates to measure activities such as chymotrypsin-like cleavage in cell extracts or suitable cellular systems. The method should identify the proteasome component and activity being assessed.
- 20S or other relevant proteasome activity assays
- Checks for fluorescence interference and nonspecific cleavage
- Protein carbonylation or aggregation measurements
- Ubiquitin-related markers where appropriate to the system
A change in a substrate-cleavage signal is not a direct percentage measure of cellular detoxification. It needs interpretation alongside assay controls and the protein-maintenance endpoints.
Stress-recovery studies
Defined particulate or other challenge models can compare treated and control cells for activity, protein damage, viability, and recovery over time. Different pathways contribute to protein turnover, and a single activity assay does not represent the whole system.
The results can guide ingredient selection for further anti-pollution research. They do not show direct pollutant removal or establish skin repair and consumer protection benefits without additional studies.
Assessing cellular protein-maintenance responses
A fluorogenic substrate assay can compare selected proteasome activities between treated and reference samples. Sample preparation, substrate selection and interference controls affect what the signal means. A greater signal does not automatically establish improved turnover of damaged proteins in intact cells.
Complementary measurements of protein damage or cellular stress can help relate enzyme activity to the research question. Viability is important because damaged cells can produce misleading changes in measured activity.
Use the findings to compare candidates or investigate a protein-maintenance mechanism. Report the concentrations and observed responses rather than a typical efficacy increase. These results alone do not establish visible rejuvenation, pollution removal or clinical anti-ageing performance.
