Cellular autophagy

What is Cellular autophagy?
Cellular autophagy testing examines how cosmetic ingredients affect the breakdown and recycling of cellular components. LC3, p62, imaging, and lysosomal measurements help characterize pathway activity, with flux analysis needed to distinguish increased turnover from blocked degradation.
Why do you need this service?
Use autophagy studies to investigate cellular material turnover under baseline or defined stress conditions. Complementary flux and damage measurements help determine whether marker changes reflect useful turnover or impaired degradation.

Cellular autophagy
Autophagy transports cellular material to lysosomes for degradation and recycling. Cosmetic research can investigate this process in skin cells under baseline or defined stress conditions. It should not be described as direct proof that an ingredient removes environmental pollutants.
Pathway measurements
Labs expose keratinocytes or fibroblasts to the test material and use complementary methods to investigate autophagy.
- LC3 puncta imaging and LC3-II measurements
- p62 protein analysis
- Lysosomal activity measurements, including suitable cathepsin assays
- Mitochondrial quality-control endpoints where relevant
An accumulation of LC3-II or autophagosomes may indicate increased formation or impaired breakdown. Measuring autophagic flux with suitable controls is therefore important. Changes in p62 also need interpretation alongside its production and degradation.
Functional and development interpretation
Concentration-response and time-course studies can compare the ingredient with appropriate reference conditions. Stress and viability measurements help establish whether the response accompanies reduced damage or cellular injury.
Reports should identify which stage of the pathway was measured and whether turnover was demonstrated. Findings can inform research on cellular maintenance under pollutant stress, but do not establish generalized detoxification, skin renewal, or consumer anti-pollution benefits on their own.
Applications of cellular autophagy studies
Comparing pathway responses
LC3 imaging, LC3-II and p62 measurements can investigate autophagy in keratinocytes or fibroblasts. More autophagosomes may indicate increased formation or blocked breakdown. Suitable turnover controls are needed before reporting increased autophagic flux, and p62 production must be considered alongside degradation.
Studying pollutant-related stress
A specified challenge can test whether pathway changes accompany reduced cellular damage. Lysosomal activity, viability and appropriate reference conditions add context. The results can guide ingredient research but do not establish direct removal of environmental toxins, generalized skin detoxification or a consumer anti-pollution benefit.
