Eye Irritation Testing

What is Eye Irritation Testing?
Defined ocular hazard endpoints using suitable laboratory methods.
Why do you need this service?
Use eye-irritation methods appropriate to the material, hazard question and applicable framework. Laboratory hazard classification and human eye-area tolerance studies are different activities.

Eye irritation testing
Ocular-hazard assessment is relevant to eye-area cosmetics and products that may accidentally enter the eye. The method must suit the formulation and the required endpoint, such as identifying materials not requiring classification or evaluating more serious eye damage.
Available laboratory approaches
Reconstructed human cornea-like epithelium models, including EpiOcular, measure tissue viability after exposure. Methods within OECD Test Guideline 492 have a defined applicability domain and interpretation scheme.
- BCOP measures opacity and permeability in isolated bovine corneas obtained from slaughterhouse material.
- HET-CAM examines effects on the chorioallantoic membrane of a fertilized hen’s egg and may contribute supporting screening data.
- Other cell-based screening methods may help compare formulations during development.
These approaches are not interchangeable. Some use animal-derived tissues even though they do not involve testing on live animals, and their regulatory roles differ.
Interpretation and documentation
A report should identify the method, controls, sample preparation, measured responses and limits of the conclusion. Additional evidence or a combined testing strategy may be needed when one method cannot resolve the hazard category. Laboratory findings support safety evaluation but do not alone substantiate claims such as ophthalmologist tested or suitable for contact-lens wearers.
Applications of eye irritation testing
Selecting a suitable model
Reconstructed human cornea-like epithelium methods can assess defined viability-based outcomes under their method-specific criteria. Acceptance criteria should follow the selected method and material applicability. Suitability, reference responses and interference must be checked for the test material.
Keeping endpoints distinct
BCOP uses isolated bovine corneas to measure opacity and permeability. It cannot assess conjunctival redness or chemosis in an isolated cornea. A laboratory eye-hazard result does not alone establish tolerance of a finished mascara or eye cream in human use; that question needs an appropriately safety-reviewed clinical scope.
