Skin Sensitisation Assessment

What is Skin Sensitisation Assessment?
Complementary evidence on sensitisation potential, with scope and exposure considered.
Why do you need this service?
Use skin-sensitisation assessment to combine suitable evidence across sensitisation key events. Method applicability, ingredient identity and expected exposure guide the assessment rather than a negative result in one assay.

Creative Bioarray provides in vitro and ex vivo cosmetic efficacy and safety assays using skin models, alongside broader cell and preclinical research services.Skin sensitization assessment
Sensitization is the induction of an immune response that can cause allergic contact dermatitis on subsequent exposure. It differs from irritation, which can occur without immune sensitization. Assessment is often ingredient-focused because mixtures can fall outside individual methods’ applicability domains.
Complementary non-animal methods
- DPRA examines reactivity with model peptides as a proxy for protein binding.
- KeratinoSens examines keratinocyte activation associated with the sensitization pathway.
- h-CLAT measures markers of dendritic-cell activation.
These methods address different key events. Defined approaches or integrated assessments combine suitable results with other information to support hazard identification and, where justified, potency assessment. A negative result in one assay does not establish that a product cannot cause allergy.
Evidence for the safety assessment
The study package should record identity, purity, test concentrations, controls, assay validity and uncertainties. Solubility, cytotoxicity and mixture composition can constrain interpretation. Existing toxicological data and intended exposure remain important when deciding whether use is acceptable.
Human repeated-insult patch tests are clinical studies, not substitutes for prior hazard assessment. Photosensitization also requires a separate evaluation when relevant. Reports should not promise a no-allergy guarantee or derive a universal safe threshold from these assays alone.
Skin sensitization and allergenicity assessment
Skin sensitization is a safety question, even when studied alongside efficacy mechanisms. No single biomarker captures the entire process. An assessment should distinguish validated methods used within their applicability domains from exploratory immune-response assays.
Inflammatory signals
Cytokine profile studies characterize immune signaling after exposure. Inflammatory mediator measurements provide related mechanistic information, but inflammatory changes are not specific to sensitization.
Early sensitization events
Dendritic-cell activation testing addresses an immune-cell response involved in sensitization. Some established methods use surrogate cell lines rather than actual epidermal Langerhans cells. Protein binding assessment examines chemical reactivity with peptides or proteins, an early molecular event for many sensitizers.
Adaptive immune responses
T-cell response studies investigate the adaptive immune component of delayed hypersensitivity. These studies are more specialized and should not be described as universally validated screening tests.
Interpretation combines relevant evidence, material properties, exposure considerations, and method limitations. Negative findings from one assay do not establish absence of allergy risk for all users.
4 subcategories of Skin Sensitisation Assessment services
Combining sensitisation evidence
DPRA, KeratinoSens and h-CLAT address different events in the sensitisation pathway. Protein-binding methods examine an early molecular event for many sensitising chemicals, while keratinocyte and dendritic-cell activation methods examine other parts of the pathway. Select methods according to material properties and applicability, with existing toxicology and exposure information available to the assessor. Mixtures, finished formulations and poorly soluble materials can require particular care in interpretation.
For cell-activation work such as h-CLAT, viability and interference checks help assess whether marker measurements are reliable. Report validity and limitations for the method actually performed rather than relying on an unspecified published study.
Cytokine, mediator or specialised T-cell studies can investigate mechanisms, but general inflammation is not specific to sensitisation. A useful assessment explains concordant and conflicting evidence, analytical limitations and exposure relevance.
Defined approaches or an integrated assessment can support hazard conclusions where justified. They do not guarantee that a product cannot cause allergy. Human tolerance testing is a separate clinical scope. Repeated-insult patch studies require prior safety justification and are not a substitute for hazard assessment or a routine screen of unknown hazards.
Hypoallergenic wording needs its own evidence review and cannot promise absence of allergic reactions in all users.
