Definition

Franz cell diffusion testing

A Franz cell has a donor compartment above a membrane and a receptor compartment below it. The test formulation is applied to the donor side, and receptor samples are collected over time to quantify transfer of a selected analyte.

Study design

Biological skin and synthetic membranes answer different questions. Excised skin can support dermal permeation assessment when integrity and handling are controlled. A synthetic membrane may be useful for comparing release from formulations, but does not automatically simulate biological absorption.

  • Defined application dose and exposure conditions
  • Controlled temperature and receptor mixing
  • A receptor medium that supports analyte recovery and suitable sink conditions
  • Timed sampling with appropriate volume corrections
  • A selective analytical method and documented recovery

Outputs and limitations

Results may include cumulative amount transferred, flux and comparisons between formulation variants. Skin retention or layer-specific concentration requires additional extraction or separation work and is not a routine consequence of receptor sampling.

The method can help compare carriers or investigate exposure, but greater receptor recovery does not necessarily mean better cosmetic performance. Franz cell data alone cannot establish dermal targeting, follicular delivery or clinical efficacy. Interpretation should state the membrane model and distinguish release, skin retention and permeation.

Applications

Interpreting diffusion-cell comparisons

A skin preparation can support permeation work, while a suitable synthetic membrane may support release testing without reproducing the skin barrier. Donor conditions, receptor medium and sampling affect what can be compared and should be documented.

Receptor-fluid analysis describes transferred amounts or flux. Skin retention and depth distribution require appropriate tissue sampling or additional analysis; they are not obtained automatically from the receptor profile.

Comparative cosmetic work needs a question-specific design and interpretation. Drug bioequivalence requirements should not be presented as a general cosmetic market-authorisation pathway. Permeation findings also do not establish efficacy or safe use alone.

Learn more about Skin Absorption Studies

Choose cosmetic penetration and delivery studies by your question: skin uptake, layer distribution, comparative delivery or release over time. Explore another Skin Absorption Studies page on cosmetic-labs.com.

Ingredient Release-Rate Studies

Measure ingredient release over time from a cosmetic formula or carrier, with release profiles kept distinct from duration of clinical product effects.

Skin absorption profiling

Measure ingredient amounts on the surface, retained in skin and permeating through it, with recovery and mass balance reported for the study conditions.

Tape stripping analysis

Measure material recovered from successive stratum corneum tape strips to compare superficial deposition, without inferring exact depths or dermal uptake.

Skin layer distribution

Skin layer distribution services for cosmetic formulas. Tape stripping, HPLC, and confocal microscopy map ingredient concentration from surface to dermis.

Active delivery assessment

Compare ingredient availability or localisation from carriers, defining the delivery question separately from absorption and distribution studies.

Franz cell diffusion testing

Measure ingredient transfer across a selected membrane with Franz diffusion cells, specifying whether the study addresses skin permeation or release.
