SOD/Catalase activation

What is SOD/Catalase activation?
SOD/catalase activation testing assesses how an ingredient affects the activity of two antioxidant enzymes in skin-cell models. Superoxide dismutase converts superoxide to hydrogen peroxide, and catalase decomposes hydrogen peroxide. Activity measurements are distinct from enzyme expression or direct chemical scavenging.
Why do you need this service?
Use SOD and catalase measurements to compare antioxidant-enzyme responses under defined conditions. Enzyme activity, protein abundance and gene expression provide complementary information and should be reported distinctly.

SOD and catalase activity testing
Superoxide dismutase (SOD) and catalase contribute to cellular control of oxidants through different reactions. Testing examines whether treatment changes their activity under basal conditions or during a defined oxidative challenge.
Enzyme activity measurements
SOD activity can be assessed through inhibition of a superoxide-dependent reaction, including methods using a xanthine oxidase system. Catalase assays follow hydrogen peroxide decomposition. Test-material interference must be considered, particularly when a compound directly scavenges oxidants or affects the detection reaction.
- Activity comparisons between treated, vehicle-control, and challenged cells
- Normalization to protein content or another appropriate reference
- Concentration-response and time-course measurements
- Viability controls and assay-interference checks
Expression and functional context
Protein or gene-expression analyses can help determine whether altered activity accompanies changes in enzyme abundance. Activity alone does not establish increased enzyme production. Likewise, increased expression may reflect a stress response rather than improved protection.
Damage markers and cellular function measurements can test whether the observed enzyme response accompanies protection in the model. These data complement direct antioxidant screens, but neither type is universally more valuable: the appropriate endpoint depends on the proposed mechanism.
Applications of SOD and catalase studies
Measuring enzyme activity
Suitable assays can examine superoxide-dependent reactions for SOD and hydrogen peroxide decomposition for catalase. Normalize to an appropriate cell or protein reference and check whether the test material directly affects the detection chemistry. A chemical scavenger can alter an assay without inducing an enzyme.
Investigating protection
Protein and transcript measurements can help explain activity changes, while damage and viability endpoints test their functional context. RT-PCR cannot by itself demonstrate catalytic activation. Increased enzyme expression can also be a stress response; neither a fixed percentage increase nor a particular assay duration establishes anti-aging protection.
