Definition

Receptor binding

Receptor binding studies investigate a specific molecular interaction proposed to explain ingredient activity. They can establish binding under defined assay conditions, but binding alone does not show receptor activation, inhibition, or a visible skin benefit.

Affinity and selectivity measurements

Depending on the target, labs may use radioligand binding, fluorescence polarization, or other suitable methods. Dissociation constants describe binding affinity. An IC50 from a competition assay is an assay-dependent inhibition measure and should not be treated as interchangeable with a dissociation constant.

  • Competitive binding with a known ligand
  • Selectivity across a relevant receptor panel
  • Association and dissociation rates
  • Investigation of possible allosteric interactions

Functional signaling studies

Follow-up experiments examine what happens after receptor engagement. Endpoints may include cAMP or cGMP levels, calcium flux, protein phosphorylation, or transcriptional activity, depending on the receptor and model.

Comparing binding and functional results helps distinguish direct receptor effects from indirect cellular changes. Reports should state the tested target, controls, concentrations, and limitations. This evidence informs mechanism-of-action research, while clinical efficacy and the suitability of product claims require additional assessment.

Applications

Applications of receptor binding studies

Testing a target hypothesis

A study can investigate interaction with a specified receptor using an appropriate ligand and binding method. Report the target, sample preparation, controls and assay conditions. An IC50 from a competition assay depends on that assay; it is not interchangeable with a dissociation constant. For a TGF-β or PDGF signaling hypothesis, identify the receptor and distinguish direct binding from downstream pathway responses.

Relating binding to response

PPAR or other target studies can compare selectivity and investigate downstream signaling in a relevant cell system. Affinity alone neither identifies a generally effective cosmetic concentration nor proves barrier repair or wrinkle reduction. Functional and product-use evidence must address those outcomes separately.
