## What is Human Tolerance Testing?

Clinical safety assessment examines cosmetic tolerance in human participants under defined protocols and clinical supervision. Patch and use tests, dermatologist or ophthalmologist assessments and comedogenicity studies provide evidence for specific tolerance claims, alongside relevant preclinical safety information.

Human-participant tolerance studies only. In-vitro phototoxicity belongs under Laboratory Hazard Testing. Hypoallergenic evidence review is a sibling, not automatically a human tolerance study.

### Why do you need this service?
Human tolerance studies evaluate reactions under defined patch or use conditions after appropriate safety review. Study design depends on the product, application site and question; no result guarantees tolerance for every user.

Definition
## Clinical safety assessment
Clinical assessments document reactions observed when a cosmetic is used under specified conditions. They complement the wider safety evaluation and do not prove that a product is risk-free for every user. Prior safety review, informed consent, appropriate oversight and adverse-event management are essential to human studies.

### Repeat-insult patch testing
[Repeat-insult patch testing](https://cosmetic-labs.com/services/in-vitro-testing/cosmetic-safety-testing/safety/patch-testing) uses induction, rest and challenge phases to assess sensitisation potential while recording irritation. It differs from a single patch test and does not reproduce normal use exactly.

### Dermatologist supervision
A [dermatologist-supervised tolerance assessment](https://cosmetic-labs.com/services/in-vitro-testing/cosmetic-safety-testing/safety/dermatologist) records and interprets skin signs and participant symptoms during a specified application study. A documentary review alone is not equivalent to a supervised use test.

### Eye-area tolerance
[Ophthalmologist testing](https://cosmetic-labs.com/services/in-vitro-testing/cosmetic-safety-testing/safety/ophthalmologist) evaluates tolerance of eye-area cosmetics under medically supervised use conditions, with attention to symptoms and relevant ocular findings.

### Light-related toxicity
[In-vitro phototoxicity testing](https://cosmetic-labs.com/services/in-vitro-testing/cosmetic-safety-testing/safety-testing/3t3-nru-phototoxicity-testing) compares cellular responses with and without light exposure. Although linked from this group, it is not a clinical volunteer study and does not assess photoallergy.

### Comedogenicity
[Comedogenicity assessment](https://cosmetic-labs.com/services/in-vitro-testing/cosmetic-safety-testing/safety/comedogenicity) investigates pore-clogging potential through suitable human studies to support a defined non-comedogenic claim. It does not guarantee that no user will develop a breakout.

[Preclinical safety testing](https://cosmetic-labs.com/services/in-vitro-testing/cosmetic-safety-testing/safety-testing) supplies complementary evidence used in planning an appropriate safety programme. The mix of studies depends on the formulation, intended use and applicable market requirements.

Applications
## Choose the human tolerance question
Patch and repeat-insult designs assess specified responses under controlled exposure. Non-comedogenic claim studies examine comedone-related outcomes, while ophthalmologist- or dermatologist-supervised use studies assess tolerance relevant to their application sites and recruited populations.

These designs have different purposes. A general patch result does not establish ocular tolerance, absence of comedogenicity or freedom from allergy. Participant selection, ethical review and sufficient prior safety information are needed before exposure.

Reports should document application, reactions, adverse events and limitations. Sensitive-skin suitability requires relevant recruitment and outcomes, not a universal pass score. Tolerance findings contribute to a broader safety assessment without replacing toxicological review.
