Dendritic-Cell Activation Testing | Cosmetic Labs

## What is Dendritic-Cell Activation Testing?

Assess activation markers in specified immune-cell models relevant to skin sensitisation. Surrogate cell-line assays, including h-CLAT where appropriate, assess markers such as CD86 and CD54 rather than directly establishing activation of epidermal Langerhans cells.

### Why do you need this service?
Immune-cell activation studies contribute to sensitisation assessment using the cell system and prediction criteria of the selected method. Surrogate cell lines should not be described as epidermal Langerhans cells, and results require integrated interpretation.

Definition
## Dendritic-cell activation and Langerhans-related responses
Langerhans cells are antigen-presenting cells in the epidermis. Their role in sensitization can be investigated directly in specialized models or indirectly through validated dendritic-cell activation methods. The exact cell system should be stated.

### Surrogate activation assays
For example, h-CLAT uses THP-1 cells and flow cytometry to measure CD86 and CD54 expression after exposure. Other methods use different cell systems and readouts. These are not interchangeable protocols, and THP-1 or U937 cells should not be described as cultured Langerhans cells.

Method-defined concentration selection and exposure conditions
- Marker expression relative to suitable controls
- Concurrent viability assessment
- Assay-specific prediction criteria and applicability checks

### Applications and limits
Results address a defined event in the sensitization pathway. Interpretation depends on solubility, cytotoxicity, analytical interference, and the method’s suitability for the material. Complex extracts and finished formulations may need particular scrutiny.

Activation data can contribute to an integrated sensitization assessment. One marker threshold is not applicable to every assay, and a negative result does not alone establish product safety or absence of allergy risk.

Applications
## Select the activation method
Methods addressing dendritic-cell activation are described under OECD Test Guideline 442E. For example, h-CLAT uses THP-1 cells and flow cytometry to assess CD86 and CD54. These cells are not cultured epidermal Langerhans cells, and other assays use different systems and readouts.

Apply the method-specific concentration selection, exposure, viability and prediction criteria. Solubility, cytotoxicity and analytical interference can limit interpretation, particularly for complex extracts and formulations. There is no generic marker threshold that applies to all activation assays.

### Integrate the result
Reports should identify the method, cell system, controls and applicability limits. Activation findings contribute to a broader sensitisation assessment rather than establishing overall product safety, guaranteed regulatory acceptance or absence of allergy risk.
