Skin Irritation Testing | Cosmetic Labs

## What is Skin Irritation Testing?

Laboratory irritation endpoints using an appropriate model.

### Why do you need this service?
Preclinical irritation studies use suitable tissue models to investigate local irritation potential. They differ from sensitisation assessment and human patch testing, and no allowable allergy rate defines a general cosmetic safety pass.

Definition
## Preclinical skin irritation testing
Skin irritation is a local inflammatory response to tissue damage and is different from an allergic sensitization response. In this preclinical service, laboratories use suitable tissue models to investigate irritation potential before considering human exposure.

### Reconstructed epidermis methods
Validated reconstructed human epidermis methods, including those covered by OECD Test Guideline 439, expose tissue to a test material under defined conditions. Tissue viability is measured after the specified exposure and recovery period and interpreted using the method’s prediction model.

Selection of a method appropriate to the sample’s physical form
- Controlled application, exposure and rinsing
- Positive and negative controls and tissue-quality checks
- Checks for interference with the viability measurement

### Using the findings
The report should state the tested material, dilution if any, method applicability and observed response. Results can inform ingredient selection, reformulation and a broader safety assessment, but do not guarantee that a finished product will be tolerated by every user.

Human patch tests, repeated-use studies and repeated-insult protocols are separate clinical investigations. They should not be treated as interchangeable with a laboratory irritation assay or as a routine way to screen unknown hazards on volunteers.

Applications
## Investigating skin irritation potential
Reconstructed human epidermis methods, including applicable OECD TG 439 methods, expose tissue under defined conditions and interpret viability using the method’s prediction model. These are tissue assays, not patch tests on volunteers.

Method selection should account for physical form, dilution and analytical interference. Positive and negative controls and tissue-quality checks support interpretation. Additional mechanistic or barrier measurements have different purposes and should not be presented as interchangeable validated irritation endpoints.

Results can guide reformulation and the broader safety review. BCOP addresses an ocular endpoint, not skin irritation. Human patch, repeated-use and repeat-insult studies require separate safety and ethical consideration. Irritation and allergic reactions need distinct interpretation within the broader safety assessment.
