Tape stripping analysis | Cosmetic Labs

## What is Tape stripping analysis?

Tape stripping analysis removes successive portions of the stratum corneum with adhesive strips and measures material recovered on them. It can compare superficial ingredient deposition, but does not directly map deeper skin layers or provide an exact depth for each strip.

### Why do you need this service?
Tape stripping compares superficial ingredient deposition within the stratum corneum after controlled application. It can inform delivery-system research, with conclusions limited to the sampled material and method. The technique may disrupt the barrier and does not establish SPF, dermal delivery or clinical efficacy.

Definition
## Tape stripping analysis
Tape stripping investigates the outer skin barrier after controlled product application. Adhesive strips remove corneocytes and associated material for chemical analysis. It is minimally invasive on volunteers and can disrupt the barrier, so it should not be described as causing no tissue disturbance.

### Controlled sampling
Application dose, contact time, surface cleaning, tape type, pressure and removal technique affect recovery. Sequential strips do not necessarily remove equal thicknesses, so strip number alone is not an exact measure of depth.

Defined application area and exposure period
- Standardized stripping and handling
- Suitable extraction and analyte quantification
- Assessment of removed stratum corneum where depth normalization is needed
- Controls for surface residue and recovery

### What the profile can show
Results can compare superficial deposition from different vehicles or delivery systems and investigate distribution of an analyte within the stratum corneum. Human and ex vivo studies require appropriate design and handling; findings from one model should not automatically be generalized to another.

Tape stripping does not directly measure dermal concentration, systemic absorption or complete skin bioavailability. Complementary diffusion or tissue-separation methods may be needed for those endpoints. Chemical results are obtained after analysis rather than as an inherently real-time readout.

Applications
## How tape stripping analysis is used in cosmetic development

### Superficial ingredient deposition
Labs apply adhesive strips sequentially to collect corneocytes and associated material from the stratum corneum. Chemical analysis of the strips can compare deposition of retinol, vitamin C, peptides or UV filters from different formulations.

Particle size, pH and carrier systems can be compared against a defined superficial-retention objective. Tape type, pressure, application dose, exposure time and surface cleaning need control. Strip number is not an exact depth measure because successive strips may remove different amounts of material.

### Comparing delivery systems
Sampling at defined post-application times can investigate how stratum-corneum recovery changes. Comparing encapsulated and free-form ingredients can inform research on liposomes or microspheres, with conclusions limited to measured deposition in the tested conditions.

Tape stripping does not establish dermal or melanocyte-layer concentrations, systemic bioavailability, SPF or clinical efficacy. Complementary tissue-separation, diffusion or endpoint-specific efficacy studies are needed for those questions. Sampling can disrupt the barrier and should be planned accordingly.
