Skin layer distribution | Cosmetic Labs

## What is Skin layer distribution?

Ingredient localisation in defined skin compartments using suitable separation or imaging.

### Why do you need this service?
Use skin-layer distribution studies to investigate an analyte in defined skin compartments or by suitable localization methods. The intended site of action and analytical recovery guide the design. Dermal delivery is not inherently desirable for every ingredient, and distribution data do not establish visible brightening or other clinical benefits within a fixed period.

Definition
## Skin layer distribution analysis
This service focuses on location rather than only total permeation. The study defines the compartments of interest, such as stratum corneum, remaining epidermis and dermis, and determines whether the chosen method can resolve them reliably.

### Methods and controls
Tape stripping for material associated with the stratum corneum
- Physical separation or sectioning of deeper tissue compartments
- HPLC or mass-spectrometric analysis of extracted analyte
- Appropriate fluorescence or confocal imaging for localization
- Diffusion systems where distribution and permeation are studied together
Surface residue, transfer between compartments during preparation and incomplete extraction can distort the apparent profile. Imaging resolution and label behavior also limit interpretation. A Franz cell alone does not produce a layer map.

### Using distribution evidence
Findings can compare vehicles or carriers and help investigate whether retention matches a formulation objective. Some cosmetic ingredients are intended to remain near the surface, and anti-aging benefits do not universally require dermal penetration.

Reports should identify the skin model, sampled compartments, analytical recovery and timing. Results from excised skin or reconstructed tissues do not automatically reproduce human use, and a concentration at a location does not establish biological activity there. Subcutaneous distribution should not be assumed unless that compartment is actually included and measured.

Applications
## How skin-layer distribution analysis is used
Cosmetic labs investigate ingredient location within defined skin compartments, such as the stratum corneum, remaining epidermis and dermis. Subcutaneous distribution is assessed only when that compartment is included and measured.

### Comparing delivery systems
Formulators compare liposomes, nanoparticles and emulsions to investigate how a vehicle changes ingredient retention. Sampling times and tissue separation are selected for the study question. Some ingredients are intended to remain near the surface, so greater penetration is not automatically a better result.

### Layer-specific concentration data
Tape stripping can sample stratum-corneum material. Sectioning or tissue separation can support deeper compartment analysis, with HPLC, mass spectrometry or suitable imaging used to detect the ingredient. A Franz diffusion cell can support a combined permeation study but does not by itself identify the layer containing an ingredient.

Reports need controls for surface residue, transfer between compartments and analytical recovery. Layer-specific amounts can inform exposure assessment and formulation selection, but do not alone establish safe concentrations, systemic exposure, therapeutic activity or finished-product efficacy.
