Skin absorption profiling | Cosmetic Labs

## What is Skin absorption profiling?

Quantities remaining on the surface, retained in skin or permeating through it, with recovery and mass balance.

### Why do you need this service?
Use skin absorption profiling to compare retained and permeated analyte under defined exposure conditions. Model integrity, sampling, analytical recovery and relevant skin compartments determine the interpretation. The profile does not establish an exact penetration depth or delivery to the dermis within a standard time, and it does not alone prove efficacy.

Definition
## Skin absorption profiling
An absorption study asks how much of an applied substance remains on the surface, is retained within the tested skin preparation or passes through it. The definitions used for absorbed material should be stated explicitly, especially when results will inform systemic exposure estimates.

### Model and sampling plan
Excised human or other justified skin preparations may be mounted in diffusion systems. OECD Test Guideline 428 provides a framework for in vitro skin absorption studies. Synthetic-membrane release testing does not substitute for a biological skin absorption study.

Documented skin source, preparation and barrier integrity
- Application conditions representative of the intended exposure
- Collection of receptor fluid and relevant wash fractions
- Extraction of retained analyte from defined tissue compartments
- Mass-balance assessment and suitable analytical sensitivity

### Interpreting the profile
Outputs can include time-dependent permeation, retained amounts and comparisons between cream, gel or serum vehicles. Layer-specific data require appropriate separation methods. A measured fraction in receptor fluid is not a direct measure of clinical bioavailability at a biological target.

Results can inform a toxicological assessment and identify how formulation changes alter exposure. Their relevance depends on the test conditions and model, and they do not alone establish efficacy, universal regulatory acceptance or a safe use concentration.

Applications
## How skin absorption profiling is used in product development
Skin absorption profiling compares surface residue, skin-retained material and permeated amounts under defined application conditions.

### Comparing formulation vehicles
Franz cell diffusion studies can track analyte collected in receptor fluid over time. Analysis of wash fractions and extracted tissue supports recovery and mass-balance assessment. Tape stripping can add information about stratum-corneum deposition, while deeper layer measurements require appropriate tissue separation.

Labs can compare liposomes, microspheres, emulsions and penetration enhancers to investigate changes in retention and permeation. Effects of particle size or carrier choice must be measured for the tested ingredient and formulation rather than assumed from a generic percentage improvement.

### Exposure assessment and documentation
Absorption data can inform systemic exposure estimates within a toxicological assessment. Calculating a margin of safety also requires relevant toxicological inputs and explicit exposure assumptions. An absorption profile alone does not establish a safe use concentration.

Leave-on, rinse-off and sun-care studies need application conditions suited to intended use. Regulatory data needs depend on the ingredient, product category, market and assessment question. The study does not guarantee registration or FDA approval.
