Neurogenic reduction | Cosmetic Labs

## What is Neurogenic reduction?

Neurogenic reduction testing investigates whether cosmetic ingredients reduce nerve-associated inflammatory responses in laboratory models. Measurements of sensory-neuron activation, substance P, CGRP, and downstream mediators help assess proposed mechanisms relevant to reactive-skin comfort.

### Why do you need this service?
Neurogenic-response studies help compare ingredient effects on nerve-associated inflammatory signals in a laboratory model. Use them to investigate a proposed soothing mechanism before assessing comfort during human product use.

Definition
## Neurogenic reduction
In this service, neurogenic reduction refers to investigating a reduction in nerve-mediated inflammatory signaling. Sensory nerves can release mediators such as substance P and calcitonin gene-related peptide (CGRP), which participate in skin responses associated with irritation and discomfort.

### Models and measurements
Labs use sensory-neuron cultures or suitable co-culture and tissue systems to represent the pathway under investigation. Ordinary reconstructed epidermis does not reproduce the full nerve-driven response unless the relevant neuronal components are included.

Stimulus-response studies, such as an appropriate capsaicin challenge
- Neuropeptide measurement using immunoassays
- Cytokine profiling of downstream inflammatory responses
- Calcium imaging to assess neuronal activation
Controls and viability measurements help determine whether lower signaling reflects a specific effect rather than toxicity.

### Interpreting a reduction
Concentration and time-course comparisons can guide ingredient selection or formulation adjustments. Neurogenic markers are not direct measurements of burning, itching, or stinging, and their relationship to perceived comfort needs separate investigation.

The findings may support a mechanistic rationale for soothing products. They do not establish treatment of rosacea or atopic dermatitis, nor do laboratory assays alone justify a dermatologist-tested claim.

Applications
## Studying nerve-associated inflammatory responses
Select a model that can represent the sensory-neuron or neuropeptide response under investigation. Measurements may examine neuronal activation, substance P, CGRP or downstream mediators. The brief should distinguish reduced release from a change in the response to an added mediator.

Controls and viability help determine whether suppression reflects a specific response rather than nonspecific damage. Report the challenge, exposure and measured endpoint with the findings instead of assuming a typical percentage reduction or response time.

Mechanistic evidence can inform ingredient selection and a later human comfort study. It is not clinical evidence of soothing, and it does not establish treatment of rosacea or another disorder. Human challenge protocols require a separately justified clinical scope.
