## What is Senescence reduction?

Senescence reduction testing investigates how cosmetic ingredients affect markers of cellular senescence in skin models. Panels of biochemical and functional measurements help distinguish changes in senescence-associated activity from altered cell viability or proliferation.

### Why do you need this service?
Senescence studies compare multiple markers and functional responses in a defined skin-cell model. Prevention, selective loss of senescent cells and changes within surviving cells need different interpretations.

Definition
## Senescence reduction testing
Cellular senescence is a state associated with stable cell-cycle arrest and changes in cell function. Testing explores whether ingredients alter senescence-related markers in a defined skin cell model, without assuming that a marker change reverses ageing or extends lifespan.

### Senescence marker panels
Laboratories may assess senescence-associated β-galactosidase activity, p16 and p21 expression, and DNA damage responses. Multiple endpoints are useful because no single measurement establishes the complete senescent state.

SA-β-gal staining
- p16 and p21 gene or protein expression
- DNA damage response markers
- Telomere length or telomerase activity where relevant to the model
Viability and cell-number controls are essential. A lower proportion of marker-positive cells may have several explanations and should not automatically be described as rejuvenation.

### Functional follow-up
Cell-cycle analysis, proliferation assays, mitochondrial respiration and autophagy measurements can explore functional changes alongside marker results. Collagen synthesis and resistance to oxidative stress may provide additional skin-relevant endpoints.

Concentration-response testing helps identify conditions for further investigation. Reports should distinguish prevention of a senescence-associated response from effects on already senescent cells. These preclinical findings can inform product development, but visible anti-ageing or age-reversal claims require evidence beyond this assay panel.

Applications
## Characterising senescence-related changes
SA-β-galactosidase, p16, p21 and DNA-damage markers can contribute to a panel. No single marker establishes or reverses the complete state. Cell number and viability are essential because fewer marker-positive cells may result from toxicity or selective cell loss.

Study design should distinguish prevention of a response from treatment of already senescent cells. Proliferation, cell-cycle, respiration or autophagy measurements can provide functional context. ATP content alone is not ATP-production rate.

The findings can prioritise candidates and guide further research. They do not demonstrate lifespan extension, cellular rejuvenation or visible age reversal, and matrix-related changes cannot by themselves substantiate skin-firming or wrinkle-reduction claims.
