## What is Keratinocyte proliferation?

Keratinocyte proliferation testing measures how ingredients influence division of epidermal keratinocytes. Cell-cycle and viability measurements characterize a cellular renewal-related response without equating increased proliferation with complete epidermal turnover or anti-aging efficacy.

### Why do you need this service?
Keratinocyte-proliferation studies investigate cell division in epidermal models. Use direct proliferation or cell-cycle evidence with viability information rather than treating general metabolic activity as a growth rate.

Definition
## Keratinocyte proliferation
Keratinocytes form the epidermis through coordinated proliferation and differentiation. This service focuses on cell division, a narrower endpoint than the full renewal process from basal cell production to surface shedding.

### Measuring cell division
Keratinocyte cultures can be exposed to samples and assessed using DNA synthesis labeling, cell counts, or proliferation-associated markers. BrdU or other suitable incorporation methods measure DNA synthesis, while imaging can add cell-number and cell-cycle information.

Concentration-response and time-course profiles
- Viability measurements to identify toxicity
- Untreated, vehicle, and appropriate reference controls
- Separation of proliferation from changes in metabolic activity

### Renewal and tissue context
Differentiation markers, morphology, and reconstructed epidermis studies can help determine whether increased division is compatible with normal tissue organization. Faster proliferation is not inherently beneficial and does not establish accelerated turnover, smoother skin, or reduced fine lines. Fibroblast activity and collagen synthesis belong to separate endpoints. Combining proliferation with differentiation and barrier data helps interpret the response within skin-aging research.

Applications
## Assessing a cellular renewal-related response
DNA-synthesis or cell-cycle measurements can help compare ingredient effects on proliferation. Bromodeoxyuridine incorporation indicates DNA synthesis, not the time needed for complete epidermal turnover. The model and observation period affect interpretation.

MTT measures a metabolic reduction signal and cannot independently distinguish more cells from altered metabolism or survival. Complementary measurements are needed when the question is proliferation rather than general cell condition.

Use the findings to investigate a renewal-related mechanism or select candidates for tissue studies. Increased division is not inherently beneficial and does not establish complete skin renewal, clinical anti-ageing efficacy or visible smoothing.
