## What is DNA repair stimulation?

DNA repair stimulation testing investigates whether an ingredient changes removal of UV-induced DNA lesions in skin-cell models. Lesion-specific measurements over time help distinguish repair from reduced initial damage, altered cell survival, or other effects.

### Why do you need this service?
DNA-repair studies investigate whether an ingredient changes the disappearance of selected UV-induced lesions over time in skin-cell models. The design needs to separate repair from reduced initial damage and changes in survival.

Definition
## DNA repair responses after UV exposure
DNA repair testing examines recovery from defined UV-induced lesions, rather than the optical filtering measured by sunscreen assays. Treatment timing and an initial damage measurement are important for distinguishing accelerated lesion removal from prevention of lesion formation.

### Lesions and repair pathways
Assays may quantify cyclobutane pyrimidine dimers or oxidative lesions such as 8-oxoguanine. Repair-pathway markers can provide mechanistic context. Comet assays detect particular forms of damage depending on the protocol and are not universal direct measurements of all UV lesions.

Comparable UV exposure and initial lesion burden
- Serial lesion measurements after exposure
- Untreated, vehicle, and appropriate reference controls
- Cell viability and proliferation checks

### Separating prevention from repair
Pre-exposure treatment can reduce the initial UV dose reaching cells, whereas post-exposure designs more directly address recovery. A decline in lesions must also be interpreted against cell loss or replication-related effects.

Where a product supplies photolyase, the assay should evaluate the supplied enzyme and its required conditions. Human cells do not naturally use photolyase as a DNA-repair enzyme. Findings support the tested repair mechanism, not an SPF value, guaranteed clinical recovery, or prevention of UV-related disease.

Applications
## Following UV-induced lesion recovery
Choose lesion-specific measurements and an observation schedule suited to the repair question. Compare the initial lesion burden as well as later measurements; a lower starting level can make a treatment appear to accelerate repair when it reduced exposure instead.

Viability and cell population changes matter because loss of damaged cells can reduce the measured signal. Complementary repair-related endpoints may help investigate a mechanism, but expression of a repair marker is not the same as measured lesion removal.

Use the findings to compare candidates and plan further UV-response work. They do not establish SPF, disease prevention or long-term skin-health benefits, and they cannot by themselves substantiate cumulative damage prevention in product users.
