Tight-junction proteins | Cosmetic Labs

## What is Tight-junction proteins?

Tight-junction protein testing examines how an ingredient affects the abundance and localization of proteins such as claudin-1, occludin, and ZO-1 in epidermal models. Protein analysis and functional permeability measurements provide complementary evidence of junction integrity.

### Why do you need this service?
Tight-junction studies investigate protein abundance and localisation in epidermal models. Functional permeability measurements can help determine whether a marker change corresponds to altered junction integrity.

Definition
## Tight-junction proteins and epidermal barrier function
Tight junctions regulate passage between cells in the viable epidermis. Testing investigates whether treatment changes the proteins and organization of these junctions, rather than assuming that higher expression alone establishes a stronger barrier.

### Expression and localization
Immunofluorescence imaging can show junctional distribution, and Western blotting or other protein assays can measure abundance. qPCR assesses corresponding transcripts, not protein concentrations.

Claudin-1 or claudin-4 expression and localization
- Occludin-associated junction organization
- ZO-1 distribution within junctional complexes
- Cell viability and monolayer or tissue integrity
E-cadherin may provide related cell-adhesion information, but belongs to adherens junctions rather than tight junctions.

### Functional interpretation
Transepithelial electrical resistance or suitable tracer-permeability assays can test whether molecular changes accompany altered barrier properties. Results depend on the model and should be compared with vehicle and reference controls across justified concentrations and exposure times.

These findings inform mechanistic evaluation of barrier-support products. They do not automatically prove barrier repair or satisfy regulatory requirements for every market or claim.

Applications
## Relating junction markers to function
Claudin-1, occludin and ZO-1 can provide information about junction components. The location of a protein matters as well as its abundance, so choose analytical and imaging endpoints according to the question. Expression data alone cannot establish a fully functioning junction.

Functional measurements in an appropriate model can strengthen interpretation of a proposed barrier effect. Consider viability and the state of the tissue when comparing treatments, particularly after a controlled challenge.

Use the findings for ingredient comparisons or to investigate a barrier mechanism. Human TEWL, perceived comfort and hydration remain different endpoints. Avoid assigning an expected protein increase or describing these laboratory measurements as clinical proof.
