## What is Corneodesmosome regulation?

Corneodesmosome regulation testing examines how ingredients affect the junctions that hold corneocytes together in the stratum corneum. Protein analysis, protease assays, and cohesion measurements investigate the balance between barrier integrity and normal shedding.

### Why do you need this service?
Corneodesmosome studies help investigate how ingredients affect corneocyte cohesion and shedding. Use protein, protease and functional measurements to examine the balance appropriate to the selected epidermal model.

Definition
## Corneodesmosome regulation and skin shedding
Corneodesmosomes maintain cohesion between corneocytes and are progressively degraded during desquamation. Testing assesses how treatment affects their persistence, breakdown, and associated tissue cohesion. Greater adhesion or slower breakdown is not always beneficial, since controlled shedding is part of normal barrier maintenance.

### Junctional proteins and proteases
Immunostaining and protein assays can assess desmoglein-1, desmocollin-1, and corneodesmosin. Kallikrein activity measurements investigate enzymes involved in corneodesmosome degradation. Expression, protein distribution, and enzyme activity provide distinct information.

Junctional protein abundance and localization
- Protease activity under defined pH conditions
- Concentration-response and time-course comparisons
- Viability and tissue-integrity controls

### Cohesion and barrier context
Tape-stripping protocols can assess stratum corneum cohesion in suitable tissue or human studies. Water-loss measurements and recovery after a controlled challenge may add functional context. Electrical resistance measures broader barrier properties and is not a direct measure of corneodesmosome strength.

The report should relate protein changes to cohesion or shedding endpoints actually measured. These results can inform barrier-support or exfoliation research, but cannot independently establish product safety or consumer benefit.

Applications
## Studying cohesion and desquamation
Corneodesmosin and desmosomal proteins help characterize the junctions involved in stratum-corneum cohesion. Protease measurements can add information about their processing. These endpoints address related processes and should not be treated as interchangeable measures of barrier repair.

Functional cohesion or shedding assessments can help interpret a molecular change. More adhesion is not necessarily an improvement, because normal desquamation requires controlled breakdown of the junctions. Excessive loss of cohesion can also be undesirable.

Findings can guide formulation comparisons or follow-up barrier studies. They do not independently establish improved human skin comfort, normalised shedding during use or a clinical anti-ageing benefit.
